Excel Spreadsheet Formatting & Manipulation

Here is what I need: Let's assume I have a Microsoft Excel spreadsheet that has 100,000 names on it (Master Excel File). And then let's assume that I have another separate file with 5,000 names on it (New Data File). What I need is to be able to remove the names from the New Data File that are duplicates of the ones in the Master Excel File. When those names have been removed from, the New Data File, I need to then create a final Excel file that has no duplicates (Scrubbed Data File). This Scrubbed Data File is the finalized spreadsheet of the remaining names from the New Data File. In other words, out of the 5,000 names, if 1,000 were already on the Master Excel File, then the Scrubbed Data File would produce 4,000 names. I have attached a visual representation of what I need. I would prefer to have a software where I can upload both the Master Excel File and the New Data File and it automatically creates the Scrubbed Data File that I can download. Please only apply if you already know you can do this.
